It's very unique in the mystery genre! Almost all historical mysteries are set in the UK (there's a ton of Victorian, but also Regency, Tudor-era, etc.). The ones set in the US tend to be set in the northwest in the 19th century.
I probably wouldn't start with Die Upon a Kiss simply because it's not one of the best ones. And you would miss out on the laying the foundations of the characters and stuff, though I don't think you would be confused. If you aren't going to start at the beginning, I would say the best place to start would be with either Fever Season or Sold Down the River, but unfortunately both of those are earlier in the series than Die Upon a Kiss!
But maybe you can try Die Upon a Kiss and see if you like it? If you do, you know you'll like the other books. If you think it's just okay, then you still might check out the stronger books in the series. If you don't like it at all then the series is probably not for you.
